Ενώ επιδιορθώνουν έναν αγωγό ύδρευσης, δύο υδραυλικοί από το Μπρούκλιν, ο Μάριο και ο αδελφός του Λουίτζι, μεταφέρονται μέσα από έναν μυστηριώδη σωλήνα σε έναν μαγικό κόσμο.
Όταν τα δύο αδέλφια χωριστούν, ο Μάριο ζει μία επική περιπέτεια αναζήτησης του Λουίτζι. Με τη βοήθεια του μανιταριού Τοντ και μετά την εντατική εκπαίδευση στη μάχη με την αρχηγό του Μανιταροβασιλείου Πριγκίπισσα Πιτς, ο Μάριο επιστρατεύει τις δικές του δυνάμεις και αναλαμβάνει δράση.
